Saroj Kumar Ghosh Anatomical and histological studies on the olfactory organ of riverine catfish, Eutropiichthys vacha (Hamilton, 1822) Background and Objective: The olfactory organ is of immense importance and plays a momentous role in various teleost behaviours. The structural detailed and functional aspects of different cells on the olfactory mucosa of Eutropiichthys vacha (Hamilton, 1822) were studied morphologically as well as histologically. Methodology: The gross morphology and the cellular composition of the olfactory epithelium in E. vacha were described by scanning as well as light microscopy. Results: The paired olfactory chambers placed on the dorsal part of the snout and communicated to surrounding environment by anterior and posterior nasal openings. The olfactory organ was lodged in the depression of the olfactory cavity and consisted of 32 ± 2 lamellae of various sizes that inserted into both sides of narrow midline raphe, forming an oval shaped rosette. The lamella was composed of olfactory epithelium whose surfaces contain sensory and non-sensory parts. The lateral surface and linguiform process of olfactory lamella contained sensory epithelium; where as the rest portion of the lamella was covered with non-sensory epithelium. The sensory epithelium was embossed with three types of receptor cells distinguished on the basis of architecture on their apical part bearing cilia, microvilli or rod like processes. The non-sensory areas were comprised of supporting cells and a series of mucous cells. Basal cells were confined in the deeper region of the epithelium above the basement membrane. Conclusion: Role of various cells lining the epithelia of the olfactory organ related to the mode of life and living of fish concerned.

  6. Saroj Kumar Ghosh Cellular organization of the olfactory epithelium during growth, maturation, spawning and post-spawning phases of freshwater catfish, Eutropiichthys vacha (Hamilton, 1822) (Teleostei: Siluriformes) Cytological status of olfactory epithelium during different reproductive phases of Eutropiichthys vacha (Hamilton, 1822) were described by light microscopy. The olfactory organ was composed of various lamella contained two principal layer: olfactory mucosa and central core, radiated from midline raphe. A sharp divergence existed between the mucosa of the olfactory lamella and raphe. The olfactory mucosa was distinguished into sensory and non-sensory epithelium, which consisted of polymorphism of receptors, supporting, labyrinth, mucous and basal cells. The central core was made up of loosely disposed connective tissue, nerve fibres and blood capillaries. A well-developed basement membrane was clearly distinguishable in between the olfactory epithelium and central core. The seasonal changes of the olfactory mucosa during growth, maturation, spawning and post-spawning phases was characterized by considering the tallness and architecture of the receptor cells as main criteria along with commute cellular features and staining patterns of different cell types. The structural components of the olfactory epithelium were correlated with the olfaction in the life of fish concerned

Soumen Roy, Korak Kanti Chaki, Tapas Chandra Nag & Kamales Kumar Misra Ultrastructure of gametogenesis in the ovotestis of an estuarine pulmonate slug, Onchidium tigrinum (Stoliczka, 1869) ABSTRACT The pulmonate slug Onchidium tigrinum (Stoliczka, 1869) is an estuarine protandrous gastropod. Transmission electron microscopy of both the gonadal and somatic cell populations of the ovotestis of the slug is documented. The acini of smaller slugs are comprised of developing spermatogenic cells and three to four small ill-developed oocytes. Details of the microscopic structures of Sertoli cells, interacinar cells and acinar boundary are described in-depth, revealing their secretory function. Sertoli cells are more numerous in the ovotestes of smaller slugs than in those of larger slugs. Tunnelling nanotubes of 200–400 nm in diameter are described for the first time in the Sertoli cells of molluscan ovotestis. These nanotubes may help to supply various cellular materials into distantly developing spermatogenic cells. The acini of larger slugs possess 2–3 mature oocytes along with a few spermatogenic cells. Sertoli cells, interacinar cells and spermatogonial cells are fewer in number in the acini of the ovotestis of larger individuals establishing the predominance of oogenesis in this phase of life. The number of oocytes per acinus is analysed in relation to the habitat of the pulmonates.

  14. Ghosh, S. K., B. Pan and P. Chakrabarti Structural organization and functional aspects of the olfactory epithelium of red piranha, Pygocentrus nattereri (Kner, 1858) Olfaction plays a momentous role in various life activities of fish. The olfactory organ of teleost is the only vertebrate organ in which nerve cells are directly exposed to the aquatic ecosystem. In fishes the receptor cells of the sensory epithelium are stimulated when they come into contact with certain chemicals carried in water. The present study describes the distribution and characterization of various cells lining the olfactory epithelium of Pygocentrus nattereri by light as well as scanning and transmission electron microscopy. The oval shaped olfactory rosette contains 28 ± 2 primary lamellae on each side of the median raphe, lodges on the floor of olfactory chamber. The olfactory chamber communicates externally by anterior and posterior nasal openings. The olfactory lamellae are composed of sensory and non-sensory epithelium in the different regions. The sensory epithelium is confined in the upper half of the lamella including tongue shaped area while the non-sensory epithelium is observed resting portion of the lamella. The sensory epithelium contains morphologically distinct ciliated and microvillous receptor cells, supporting cells and basal cells. The non-sensory epithelium is comprised of labyrinth cells, mucous cells and stratified epithelial cells. According to transmission electron microscopical observation elongated rod shaped structure emerging out from dendrite end of the receptor cells in the free epithelial surface. The supporting cells have lobular nucleus with clearly seen electron dense nucleolus. The apex of the ciliated supporting cells is broad and provided with plenty of kinocilia. Abundant electron lucent granules and microvesicles are often found in the cytoplasm of labyrinth cells. Basal cells are provided with oval nucleus and contains small number of secretory granules. Mucous cells are characterized with basally placed nuclei and filled up with granules. The functional importance of different cells lining the olfactory mucosa is correlated with respect to ecological habits of fish concerned.

  13. Saroj Kumar Ghosh and Padmanabha Chakrabarti Morphological and histochemical studies on the olfactory rosette of bagrid catfish, Rita kuturnee (Sykes, 1839) In the present study, the functional anatomy, histological features and histochemical localization of two enzymes viz., alkaline phosphatase (ALPase) and adenosine triphosphatase (ATPase) of the olfactory epithelium in Rita kuturnee were described. The structural organization of the olfactory epithelium was studied by using the histological and histochemical techniques. The morpho-anatomical study revealed that the paired olfactory chambers were situated at the dorsal-lateral sides of the snout, which was communicated outside by an anterior and posterior nasal openings. The elongated olfactory rosette lied at the bottom of chamber and supported with 54-56 lamellae on either side of the central narrow raphe. The histological analysis characterized the olfactory epithelium of lamellae which was made up of receptor, flagellated supporting, non-flagellated supporting, labyrinth, mucous and basal cells. Intense alkaline phosphatase and adenosine triphosphatase activity were evidenced in the receptor cells, supporting cells, basal cells and also in blood cells of the central core. Various cells lining the olfactory epithelium were correlated with the functional significance of the fish was concerned.
  14. Saroj Kumar Ghosh and Padmanabha Chakrabarti Comparative studies on histology and histochemistry of pancreas between Labeo calbasu (Hamilton, 1822) and Mystus gulio (Hamilton, 1822) The cellular architecture and histochemical nature of the enzyme secretory acinar cells of pancreas have been investigated in two species of fish, Labeo calbasu and Mystus gulio having different feeding habits. Histological analysis illustrates that the exocrine pancreatic tissues are scattered within the hepatic parenchyma as hepatopancreas and spleen as spleenpancreas in L. calbasu. The structural analysis also shows that exocrine as well as endocrine pancreatic tissues are also diffused in the adipose tissues in between the intestinal coiling of L. calbasu. In M. gulio the discrete pancreatic tissue is attached to the outer wall of the stomach and formed cluster of acini interspersed with small areas of islet of Langerhans and blood vessels. The exocrine pancreatic acinar cells are provided with conspicuous nuclei and densely packed acidophilic zymogen granules. The endocrine component of pancreas, islet of Langerhans is encircled by a thin connective tissue and comprised of α, β and δ cells. Different cell types in the islets are differentiated based mainly upon their shape, position, staining intensity and density of the cytoplasmic secretory granules. α cells contain sparse cytoplasm and densely stained nuclei, are generally located to the periphery of the islet. Aldehyde fuchsin positive β cells bear spherical nucleus and granular cytoplasm, are in the center of islet of Langerhans contacted with blood vessels. δ cells are few in number with light stained nuclei disperse in the pancreatic islet. Histochemical detection displays that the zymogen granules of exocrine acinar cells showing variegated intensities of tryptophan reaction, the precursor of numerous pancreatic enzymes which probably related to the food and feeding habits of the fishes concerned. The zymogen granules are larger in size and heavily deposited in the acinar cells of hepatopancreas of L. calbasu in comparison to M. gulio may be correlated with the herbivorous feeding habit in respect to secretion of digestive enzymes.
